Histology


Before you begin this section in lab, please read the page on the Use of the Microscope. This will allow you to get the best view of the specimens and help us take care of our investment.


Tissue Preparation

In order to prepare tissue for viewing under the light microscope, the following steps are usually taken:

·  A small tissue sample is taken and "fixed" by infiltrating it with a preservative. This fluid is usually water-based.

·  The fixed material is gradually passed stepwise through a series of solutions, each with increasing concentrations of alcohol.

·  The material, impregnated with alcohol, is gradually passed through a series of solutions with increasing amounts of organic solvent.

·  Paraffin is substituted for the organic solvent in a stepwise manner.

·  Paraffin blocks are solidified, each about the size of a mini ice cube.

·  Serial sections of the tissue in paraffin are made using a microtome.

·  Strips of serial sections are placed on a slide and heated slightly to help them adhere.

·  Slides with specimens are placed in stains, selected to show prominent characteristics of the sample.

·  Mounting medium is place on the slide and a coverslip is applied.
